Calpain system is thought to play an important role in post-mortem muscle proteolysis and meat tenderization processes. In skeletal muscle, the calpain system consist of the ubiquitously expressed µ-calpain and m-calpain, calpastatin inhibits both µ-calpain and m-calpain. Expression of CAST (Calpastatin gene) , CAPN1 (calpain 1 gene) and CAPN3 (calpain 3 gene) was assessed in longissiumus dorsi muscle divergent for shear force, and CAST, CAPN1 and CAPN3 gene polymorphism was been reported to be associated with meat tenderness. In this study, 3 CAPN1 markers explained variations of Warner-Bratzler shear force in Hanwoo. A significate marginal mean difference in genotypes of g.6545C>T (P = 0.046) was found in moisture with additive effects. A Significant effect (P≦0.015) of a single nucleotide polymorphism in the CANP1 gene on calcium content was observed. Samples carring CAST g.76,872AA genotype showed higher autolyzed µ-calpain activity 24 and 72 hour post-mortem. Higher CAST and CAPN3 expression was found in LD with high shear force (P<0.2), confirming a direct role for calpastatin but not for calpain 3 in meat tenderization.
