Obesity is a risk factor for chronic diseases. Therefore, prevention of obesity becomes crucial in public health. Studies showed that green tea (Camellia sinensis, GT) possesses significant anti-obesity,anti-diabetes, and cardio-protective abilities. High-fat-fed obese mice treated with 0.32% Epigallocatechin-3-gallate (EGCG) for 6 weeks can decrease body weight (BW) gain about 44% and increase fecal lipid content about 29.4% compared to controls. In vitro, EGCG inhibited pancreatic lipase (PL) with 50% inhibition at 7.5 µmol/l. Green tea polyphenol (GTP: 0.8, 1.6, 3.2 g/L) treatment attenuated the visceral adipose tissue (VAT) accumulation, hypoadiponectinemia, and the decreased mRNA level of adiponectin while increased PPARγ and phosphorylation of erk1/2 expression and decreased phosphorylation of PPARγ induced by high-fat-fed obese rats. VAT was also cultured with high glucose to mimic the hyperglycemia condition in vitro. The results were similar with in vivo. Obese rats treated with GT (12 weeks/5 days/week; 500 mg/kg BW) showed a significant reduction in hyperlipidemia, fat synthesis, BW, and fat depots through AMPK activation. It is concluded that green tea could decrease BW gain and VAT accumulation via erk1/2-PPARγ-adiponectin or AMPK pathway.
