Doxorubicin-loaded fucoidan nanoparticles for drug delivery in cancer treatment

摘  要: Doxorubicin (DOX) is a widely used chemotherapeutic agent. Nanoparticles (NPs) achieve drug delivery. Fucoidan (Fu) is a naturally occurring sulfated polysaccharide extracted from marine brown seaweeds. It has a wide spectrum of activity in biological systems. The aim of this study is to investigate DOX-Fu NPs for drug delivery and cancer treatment. The first study was to design self-organized acetylated fucoidan (AcFu) nanoparticles. DOX-loaded AcFu nanoparticles. Combine immunotherapy and chemotherapy in a single system. The DOX loading efficacy and content were 71.1% and 3.6%, respectively. DOX-AcFu NPs exhibited first-order drug release behavior for 5 days. AcFu NPs treated Raw 264.7 macrophages overexpressed various anti-tumor cytokines. The ability of DOX–AcFu to suppress drug efflux was revealed by confocal microscope images and FACS analysis in multidrug resistance (MDR) cells. The second study demonstrates the multifunctional DOX-loaded Fu capped gold nanoparticles (DOX- loaded Fu AuNPs) for drug delivery and photoacoustic imaging (PAI). To reduce the use of chemical and toxic waste to the environment, use Fu as a reducing agent for the green synthesis of AuNPs. The release of DOX from DOX- loaded Fu AuNPs was greater in acidic pH 4.5 than in neutral pH 7.4. The in vitro cytotoxic effect of DOX-Fu AuNPs proved the excellent anticancer activity of DOX-Fu AuNPs against human breast cancer cells (MDA-MB-231). DOX-Fu AuNPs induced both early and late apoptosis in a concentration-dependent manner compared with untreated control cells. The ability of DOX-Fu AuNPs as a contrast agent for in vitro breast cancer imaging with PAI has been evaluated. In conclusion, these findings of the work suggested that the multifunctional DOX-Fu NPs may be a suitable carrier for drug delivery, cancer treatment and PAI.
