Antioxidant and Angiotensin-Converting Enzyme (ACE) Inhibitory Activity of Enzymatic Hydrolysates from Clam

摘  要: Marine protein hydrolysates have antioxidative and ACE inhibitory activities. Li et al. (2016) collected the samples of Sinonovacula constricta from local market. The samples were hydrolyzed with pepsin, trypsin, alcalase, flavourzyme and Actinomucor elegans T3 protease to produce Sinonovacula constricta enzyme hydrolysates. Among the proteases investigated, flavourzyme hydrolysate (45.87%) showed the highest level of degree of hydrolysis (DH) after 3h, followed by A. elegans T3 proteases (37.84%), alcalase (30.55%), pepsin (18.72%) and trypsin (15.67%). The A. elegans T3 proteases was observed to be more effective in generating small peptides with ACEinhibitory activity. The 3 kDa peptide of A. elegans T3 proteases hydrolysate showed the highest ACE-inhibitory activity with an IC50 of 0.79 mg/mL. After chromatographic separation by Sephadex G-15 gel filtration and reverse phase-high performance liquid chromatography (RP-HPLC), the potent fraction was subjected to MALDI/TOF-TOF MS/MS for identification. A novel ACEinhibitory peptide (VQY) was identified exhibiting an IC50 of 9.8 µM. Yu et al. (2018) collected the samples of R. philippinarum from commercial market and hydrolyzed with trypsin, neutrase and pepsin to produce hydrolysates by trypsin (tRPH), neutrase (nRPH) and pepsin (pRPH). The antioxidant activities of R. philippinarum hydrolysates against inhibition on linoleic acid peroxidation (91.23%), DPPH radical scavenging (68.55%) and reducing power (0.51) showed that the nRPH exhibited the strongest antioxidant activity. In addition, an ACE inhibition assay revealed that the pRPH had the highest ACE inhibitory ability (IC50 = 0.42 mg/mL). In conclusion, the razor clam hydrolysate obtained by A. elegans T3 proteases could serve as a source of functional peptides with ACE-inhibitory activity while Ruditapes philippinarum protein hydrolysates could be a promising source of natural antioxidant and ACE inhibitory.
