Periodontitis is an inflammatory disease caused by the dental plaque, also known as a biofilm of the tooth microorganisms, which release exosome to stimulate the production of pro inflammatory factors. The inflammatory mediators trigger the soft tissue disruption and
alveolar bone resorption, which is one of the main reasons for tooth loss in adults. Here, we aim to develop an in situ injectable chitosan-based hydrogel that could decline surgery times with biocompatibility, biodegradation, and bio-adhesion properties. In this study, a chitosan based hydrogel delivery system was used to be a drg carrier, containing anti-inflammatory medicine, antimicrobial medicine, or erythropoietin (EPO) to exert pharmacological effets of anti-inflammation and tissue regeneration. In result, these formulations exhibited thermosensitive and pH-dependent swelling/erosion properties. On the other hand, the biocompatibility and biodegradation properties have been confirmed in the in vivo study. Immunohistochemistry staining and micro-CT analyses indicated that the formulation could terminate the inflammation and recovered the height of the alveolar bone, and there was no toxicity both in vitro and in vivo. In conclusion, the chitosan-based hydrogel provides a great potential candidate for periodontitis treatment in the dental clinic.
