Thermosensitive Hyaluronic Acid/Pluronic F127 Complex Hydrogel for Wound Healing

摘  要: Hydrogel demonstrates superior performance when used as wound dressing, as it absorbs wound exudates, provides a moist interface on the wound for rapid injury repair and prevents pain and injury during replacement of the dressing. Complex hydrogel can improve the disadvantages of single component hydrogel. This study aims to develop a novel thermo-sensitive complex hydrogel by combining natural polymer (hyaluronic acid, HA) and synthetic polymer (pluronic F127, F127/poloxamer 407, POL). HA/F127 complex hydrogel was evaluated by the physicochemical properties, antibacterial capacity and potential application as wound dressings. HA-POL hydrogel was prepared by stirring 1% 1200 kDa HA, 4% 120 kDa HA and 5% (w/v) POL in an ice bath. HA‐POL hydrogel transformed from sol to gel at 30 ℃, close to body temperature. HA-POL hydrogels with various thicknesses (0.04-0.22 mm) were found to have better moisture absorption and air permeability capacities than Band-Aid, a typical wound covering. HA‐POL hydrogel effectively isolated skin‐wounds from Escherichia coli invasion for 5 days, and promoted fibroblast accumulation, granulation tissue formation, angiogenesis and collagen deposition in Sprague Dawley (SD) rats after fourteen days. HA-κ-C-F127 hydrogel was prepared with HA, F127 and κ-Carrageenan (κ-C) for controlled meropenem delivery. It had the thermal stability of the fabricated injectable hydrogel with tunable gelation properties. Meropenem-loaded hydrogel showed a sustained release of meropenem in phosphate buffer saline (PBS) at pH 7.4 of 96.12%, and in simulated wound fluid (SWF) at pH 6.8 of 94.73% at 24 h. It also showed high zone of inhibition (ZOI) values against Pseudomonas aeruginosa (30 mm), Staphylococcus aureus (20.66 mm), and Escherichia coli (22.66 mm) respectively. In addition, it would increase the wound closure rate (90%) in SD rats after fourteen days. These results showed that the HA/F127 complex hydrogel using alone or with κ-C loaded meropenem had the potential to promote wound healing.
