Alginate oligosaccharides (AOS) produced by enzymatic degradation show versatile physiological functions and biological activities. In this study, two thermostable alginate lyases from different sediments were investigated. First thermostable alginate lyase Aly08 was discovered from sediments at the Yellow Sea bottom, and second thermostable alginate lyase AMOR_PL7A was discovered from hot sediments at the Arctic Mid-Ocean Ridge (AMOR). Two recombinant alginate lyases were respectively expressed in E. coli. The molecular weight of Aly08 was about 35 kDa, and the specific activity was 841 U/mg. The optimal temperature and pH of Aly08 were 45 °C and 8.35, respectively. Aly08 was also a thermo-tolerant enzyme that recovered 70.8% of its initial activity following heat shock treatment for 5 min. Aly08 degraded alginates into disaccharides and trisaccharides in an endo-manner. The optimal temperature and pH of AMOR_PL7A were 65 °C and 6, respectively. AMOR_PL7A degrades alginate and poly mannuronate within a broad range of pH, temperature and salinity, and the degradation product is oligomers with DP (degree of polymerization) > 5. These findings suggest that Aly08 and AMOR_PL7A would be a potent tool for the industrial applications.
