Drug-loaded Ulvan Complex Hydrogel for Potential Wound Dressing Applications

摘  要: Wound healing is well known to be an essential and intricate process that can lead to clinical problems; hence finding an efficient approach to enhance the healing process is necessary. In this regard, polysaccharide hydrogels have been commonly used as wound dressings for their biocompatibility, biodegradability, and non-toxicity. Ulvan (U) is a natural polymer and type of sulfated polysaccharides from green seaweed. U has the potential as a wound dressing material based on its biopolymer characteristics. The aims of this study were to develop U complex hydrogel, and investigate its physical and chemical characteristics, antimicrobial ability, and the potency of U for wound dressing application. U hydrogel films (UHF) were prepared through ionic crosslinking with U (5, 7.5 and 10%) and boric acid (0.1%), with the addition of glycerol (2%) as a plasticizer. The results showed that the higher the concentration of U in UHF, the higher the value of viscosity, swelling degree, and moisture content. Besides, all hydrogel films could reduce hydroxyl radicals and inhibit gram-positive and negative bacteria (Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli, and Streptococcus epidermidis). Authors have developed novel chitosan-U (CS-U) hydrogels
incorporating cellulose nanocrystals (CNC) and epidermal growth factor (EGF) (CS-UCNC28 EGF) by the freeze-dried process. The incorporation of CNC modified the porous microstructure, and improved mechanical properties, thermal and swelling behavior. CS-U CNC revealed non-toxic behavior and excellent cell proliferation for L929 cells. Moreover, CS-UCNC-EGF hydrogels showed significantly faster-wound healing efficiency, which was associated with faster granulations tissue formation and collagen deposition. These results indicate the potential of drug-loaded Ulvan complex hydrogel as a wound dressing.
