Parkinson's disease (PD) is the most prevalent neurodegenerative movement disorder illness of the central nervous system. Both perillyl alcohol (PA) and Withania Somnifera contain anti-inflammatory and antioxidant properties and have the ability to protect neurons. The purpose of this study is to investigate the neuroprotective effects of two plant extracts on SH-SY5Y cells that have been exposed to 6-hydroxydopamine (6- OHDA) as a PD cell model and to evaluate the potential application of plant extracts for the treatment of PD. Pretreatment with PA mitigated 6-OHDA induced cytotoxicity in a dose-dependent manner that observed marked restoration of cell viability. Moreover, after treatment with PA, it significantly reduced cytochrome c immunofluorescence and attenuated mitochondrial membrane potential loss, further alluding to its therapeutic potential against mitochondrial dysfunction. SH-SY5Y cells were treated with 6-OHDA preceding or followed by treatment with the KSM-66, W. somnifera root extract. Using KSM-66 concentrations to range from 0.25 to 1 mg/m before and after treatment of the cells with 6-OHDA has resulted in increased viability of SH-SY5Y cells. KSM-66 also modulated oxidative response proteins: peroxiredoxin-I, VGF nerve growth factor inducible and vimentin proteins upon 6-OHDA pre/post treatment. Pre-treatment of SH SY5Y cells with KSM-66 decreased protein-glutathionylation levels in the cells treated with 6-OHDA. The rescue of mitochondria with 0.5 mg/mL KSM-66 extract indicated an increase in ATP levels. In conclusion, PA and KSM-66 extracts both show neuroprotective properties and may be utilized as natural active ingredients of plants in PD drugs.
