Curcumin (CUR) has anticancer and chemoprevention properties. In order to improve its low solubility in water and bioavailability, nanoparticles (NPs) are suitable for the delivery of CUR to tumors. NPs can change the pharmacokinetic properties of CUR to improve their efficiency and decrease the side effects. Hence, the report aims to prepare Cur-loaded nanoparticles by two types of nanoformulations, different layers of magnetic alginate/chitosan and the variants of poly-glycerol-malic acid dodecanedioic acid (PGMD), for breast cancer treatment. The results showed that magnetic alginate/chitosan nanoparticles (MACPs) were successfully prepared by a layer-by-layer coating of chitosan (CHI) and sodium alginate (SA) onto the surface of Fe3O4 NPs. Moreover, the release of CUR from magnetic nanoparticles (MNPs) was possible to control release rate by changing the outermost polymer and the number of layers. The PGMD polymers were synthesized by changing the molar ratio of DDA and malic acid (7:3 and 6:4); the study revealed PGMD 6:4 was more hydrophilic than PGMD 7:3. CUR encapsulated by both PGMD polymers exhibited anticancer activity on breast cancer cell lines, and there is no significant difference between the two. Furthermore, the CUR NPs presented better anticancer activity than the cells treated with CUR only, and also displayed the breast cancer cellsin apoptotic. Overall, the Cur loaded NPs provided excellent alternative strategies for breast cancer treatment, and the CUR nanoformulations used had potential in biomedicine field.
