Preparation of alginate oligosaccharides from different alginate lyases

摘  要: Alginate is composed of β-D-mannuronic acid (M) and α-L-guluronic acid (G). Alginate oligosaccharides (AOS) are favored for their excellent biocompatibility and water solubility, showing potential in industries such as food, pharmaceuticals, and biofuel production. Enzymatic methods exhibit broad substrate specificity and site-specific cleavage reactions, resulting in different end products. AlyRm3 displays a preference for high temperatures with an optimal temperature of 70°C, yielding alginate disaccharides and trisaccharides as end products. ALY07 exhibits high activity over a wide pH range (pH 5.0-11.0) and good stability at temperatures below 35°C. Its activity is independent of NaCl, and it primarily produces disaccharides. Both enzymes are endo-type alginate lyases, demonstrating potential for efficient production of low-degree AOS. The large molecular size, low water solubility, and limited bioavailability of alginate restrict its applications. Therefore, utilizing alginate lyases to degrade alginate into low-molecular-weight alginate oligosaccharides is crucial for improving bioavailability, significantly enhancing effectiveness, especially in various industrial applications.
