The effects of purple sweet potato extract on liver damage in high fat diet induced obesity mice

摘  要: In recent years, obesity has become a common global health issue. The World Health Organization defines obesity as abnormal or excessive fat accumulation. Although there are many anti-obesity drugs available, they still pose many adverse risks to the human body. Therefore, people around the world are researching the use of natural compounds to treat obesity-related symptoms. Sweet potato is a high-yield economic crop that contains anthocyanins, polysaccharides, and dietary fiber. It has hepatoprotective, antioxidant and anti-inflammatory activities. Among them, purple sweet potato (PSP) contains the most anthocyanins and its anthocyanins are more stable than those in other plants. Therefore, the purpose of this study is to investigate the effects of purple sweet potato extract on liver damage induced by high fat diet. Evidence shows that inhibiting hepatocyte apoptosis may be a potential therapy for NAFLD. In high fat diet (HFD) induced obesity mice, purple sweet potato color extract (PSPC) can restore body weight, liver inflammation indices and apoptosis rates to normal levels. It can inhibiting oxidative stress in the mice liver and promoting Sirt1 activation, thereby inhibiting p53-induced apoptosis. PSPE can significantly inhibit cleaved-caspase-3 and Bax protein levels and activity, and increase Bcl-2 levels to reduce hepatocyte apoptosis and effectively protect the liver. Polysaccharides extracted from purple sweet potato (PSPP-A) are a new type of heteropolysaccharide composed mainly of arabinose. Itsmolecular weight is 2.63 × 103 31 kDa. It can relieve abnormalities in body weight, liver index and liver damage caused by high fat diet. And achieved by inhibiting TC and TG levels and improving liver inflammation indices to prevent NAFLD induced by a HFD in C57BL/6J mice. These two results show that both the pigment and polysaccharide extracts of purple sweet potato have the potential to repair and inhibit liver damage caused by obesity. This suggests that purple sweet potato extracts have application potential for improving liver damage
