The Effects of Vitellaria Paradoxa Nut Extract on Osteoarthritis in Meniscal/Ligamentous Injury of Obesity-Induced Rat

Osteoarthritis is an arthritis disease usually happened in adulthood after age of 45. Risk factors for osteoarthritis include age, sex, obesity, heredity and diet. Obesity caused Osteoarthritis is attributed to joint loading, cartilage disintegration, bone loss, and inflammation as well. However, treatments for this disease include moderate exercise, oral medication, intra-articular injection and artificial joint displacement surgery. Vitellaria paradoxa have been used for anti-oxidative, anti-inflammatory function. The aim of study is to investigate the fact of Vitellaria paradoxa nuts extract for meniscal/ligamentous injury and high fat diet double induced-osteoarthritis. Male Wistar rats were fed 45% high-fat diet for 4 weeks to induce obesity before transecting the anterior cruciate ligament and excising a segment of the medial meniscus of the knee to initiate Osteoarthritis. Vitellaria paradoxa nuts extract was treated in three doses (111.6, 223.2, 446.4 mg/kg per day, p.o. daily) with 8 weeks high fat diet in parallel. We observed that the swelling in the Osteoarthritis knee was the alleviated and the hind paw weight distribution was rectified after feeding Vitellaria paradoxa nuts extract. The results showed that VP decreased TG and increase of HDL-C level in high dose VP group. Next, this study found that the level of antioxidant enzymes was increased in the high dose VP group; lipid peroxidation and the level of inflammation cytokines, including NO, IL-6 and TNF-α were decreased. Our study demonstrates that feeding of Vitellaria paradoxa nuts extract shall improve meniscal/ligamentous injury of obesity-induced osteoarthritis.
