The Effects of Ethanol Extract from Velvet Antler of Formosan Sambar Deer to Male Reproductive Function on High-Fat Diet-Induced Obese Rat

Formosan sambar deer (Cervus unicolor swinhoei) is an endemic subspecies in Taiwan. The velvet antler is extraordinary valuable Chinese traditional medicines and commercial functional foods with enhancing immune system and reproductive function. Obesity comes up with various disorders such as type 2 diabetes, cardiovascular disease, hyperlipidemia, coronary diseases, and also infertility. The risk of obesity appears to originate from disruption in adipose tissue function leading to a chronic inflammatory state. The purpose of this study was to investigate the potential of velvet antler ethanol extract (VAEE) for treatment of high-fat diet-induced hyperlipidemia and reproductive damage. In vivo, Male Sprague-Dawley rats were fed 45% high-fat diet for 4 weeks to induce obese model. Velvet antler ethanol extract was treated in three doses (2, 4, 8 mg/kg per day, p.o. daily) with 5 weeks high fat diet in parallel. After sacrifice, the hypothalamus, fat, testis tissues and blood samples were collected for biochemical analysis. To determine whether VAEE reduces high-fat diet induced hyperlipidemia and its reproductive dysfunction, we observed the decrease of body weight, fat mass, TG index and increase of HDL level in high dose group rats. Secondly, we found the sperm parameters such as counts, mobility was all restored while the lipid peroxidation was reduced. In conclusion, velvet antler ethanol extract beneficially ameliorates obesity and reproductive parameters in obese rats.
