Structural Characterization and Anticoagulant Activities of Sulfated Polysaccharide from Three Algae

The structural and anticoagulant activity of polysaccharides from three algae are studied. CP2-1 was isolated from the green alga Codium divaricatum. The backbone of CP2-1 was mainly composed of (1→3)-β-D-galactopyranose residues, branched by single (1→)-β-D-galactopyranose units attached to the main chain at C-4 positions. Sulfate groups were at C-4 of (1→3)-β-D-galactopyranose and C-6 of non-reducing terminal galactose residues. CP2-1 possessed a high anticoagulant activity as assessed by the APTT assays. xylofucan was isolated from the brown alga Punctaria plantaginea. The backbone of xylofucan was (1→3)-linked α-L-fucopyranose residues. Highly sulfated linear xylofucan derivatives were shown to inhibit clot formation in APTT assay. Am-SPs was isolated from the red alga Acanthophora muscoides which contains structurally heterogeneous sulfated polysaccharides (Am-SPs). Am-SPs composed of alternating 4-linked-α-galactopyranosyl units and 3-linked-β-galactopyranosyl units presenting variable sulfation, CH3 substitutions and 3,6-anhydro-α-L-galactose units and pyruvated-D-galactose residues. Am-SPs show significantly lower anticoagulation than heparin in APTT assay.
