Study on antibacterial activity and antioxidant activity of polysaccharides from Laminaria japonica

摘  要: The topic of this study is to investigate the antibacterial activity and antioxidant activity of polysaccharides from Laminaria japonica (LJPA-P). Three acidic polysaccharides LJPA-P1, LJPA-P2, and LJPA-P3 were obtained from L. japonica by sequential hot water and citric acid extraction, and fractionation by DEAE-Sepharose fast flow chromatography and serial NaCl solution elutions. Results showed LJPA-P3 exhibit remarkably high oxygen radical absorbance capacity (ORAC, 1247.22 μmol Trolox/g) and ABTS radical scavenging activity (up to 70%). Fucoidans are sulfated polysaccharides in brown algae. The fucoidans from L. japonica were depolymerized through high-pressure hydrolysis, their depolymerized products can effectively (p < 0.05) inhibit the proliferation of Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus). The results also indicated that the bactericidal pathway of depolymerized fucoidans should be through destruction of the cytomembranes and the target molecules are the membrane proteins, which could result in changed membrane fluidity and activated autophagocytosis. With these findings, polysaccharides from L. japonica represents a good source of antioxidants and antibiotic.
