Sulfated polysaccharides represses lung cancer metastasis by inhibiting VEGF and MMPs related angiogenesis and proliferation in vitro and in vivo

Lung cancer is a high mortality rate in the world. Because the initial diagnosis of lung cancer is not easy, most of the diagnoses are in the late stage of cancer. Metastasis is responsible for about 90% of cancer deaths. Therefore, sulfated polysaccharides are from brown seaweed, with a wide variety of biological activities including anti-inflammatory, anti-cancer, and anti-viral. This study aimed to investigate the anti-metastatic effect of sulfated polysaccharides on lung cancer and explore the underlying mechanism. In past studies, Sargassum fusiforme polysaccharides(SFPS) had been shown to have good anti-tumor activity. SFPS significantly inhibited the formation of blood vessels via decreasing CD31, vascular endothelial growth factor (VEGF)-A expression and tumor microvessel density (MVD). SFPS also inhibited the expression of VEGF-A in tumor cells and VEGFR2 and the apoptosis of human umbilical vein endothelial cells (HUVECs). Fucoidan has prophylactic effects on the mitigation of cachexia and the improvement of lung masses in tumor inoculated mice. These desired effects were attributed to inhibition of LLC spreading and proliferation in lung tissues. Fucoidan also down-regulated the expression of matrix metalloproteinases (MMPs), NF-κB, and VEGF. The fact is that fucoidan significantly decreased the cell viability, migration, invasion, and MMPs activities of LLC cells. In summary, these findings suggested that sulfated polysaccharides could be used as an alternative anticancer agent as they inhibited the angiogenesis, microvessel formation and lung carcinoma metastasis through down-regulating metastatic factors VEGF and MMPs.
