Quantitative analysis of human milk oligosaccharides by liquid chromatography-mass spectrometry

摘  要: Human milk oligosaccharides (HMOs) are the third most abundant nutrients in human milk, having a board range of biological functions, and thus playing an essential role in infant health. It is therefore important to quantify HMOs for academic researches or industrial applications. However, quantifying HMOs is challenging due to their complex structures and types of isomerism. On top of that, high performance liquid chromatography (HPLC) and capillary electrophoresis (CE) are commonly used to quantify HMOs, but methods are time-consuming in sample preparation and not structurally selective. To overcome these problems, researchers have applied th combination technique of liquid chromatography (LC) and mass spectrometry (MS) to quantify HMOs, such as LC-MS in selected ion monitoring (SIM) mode and ultra-high liquid chromatography (UPLC)-MS/MS in multiple reaction monitoring (MRM) mode. The methods presented in this seminar report are easy, fast, and structurally selective due to the simple sample preparation,. the analysis time within one hour, and the ability to separate and analyze different HMOs within a single run. Additionally, the above methods show good performance in linearity, sensitivity, precision, accuracy, and recovery. The results confirm that liquid chromatography coupled with mass spectrometry is an effectively quantitative tool for the analysis of HMOs.
