The therapeutic effects of Houttuynia cordata extracts on influenza A virus infected mice

Houttuynia cordata is traditionally used for the treatment of pneumonia, bronchitis, and chronic obstructive respiratory diseases. Phytochemical investigations indicated that polysaccharides and flavonoids are important bioactive constituents in H. cordata. The aim of this study was to explore the therapeutic effects of H. cordata extracts on the disease induced by H1N1. Mice were infected intranasally with influenza virus H1N1 (A/FM/1/47). H. cordata polysaccharides (HCP), flavonoids from H. cordata (HCF) and ribavirin were administered intragastrically. Survival rates, life spans, weight losses, lung indexes, histological changes, inflammatory infiltration, and inflammatory markers in the lungs were measured. In vivo experiments showed that H. cordata extracts increase in the survival rate of H1N1 infected mice and the protection from lung and intestine injury, accompanied with the reduced virus replication, and markedly decreased the concentration of pulmonary cytokines and chemokines. In vitro experiments showed that the expression of TLRs and NFκB p65 in lung wereconstrained by H. cordata extracts treatment. This study demonstrated thatpoly saccharides and flavonoids in H. cordata not only targeted viral replicationmachinery, interfered with influenza virus-related host TLR signaling, but also providesa potential clinical therapy to treat influenza A virus infection in human beings.
