Application of Anticoagulant sodium alginate sulfates and their mussel-inspired heparin-mimetic coatings

Sodium alginate sodium sulfate ( SASs ) has a chemical structure and biological activity similar to heparin. Blood clotting time tests showed that heparin-like SAS showed excellent sulfate-dependent anticoagulant activity. In addition to the use of anticoagulants, heparin mimics also show potential applications for surface modification of blood contact devices. Dopamine-grafted SAS ( DA-g-SAS ) can be coated on a polymer substrate surface in an alkaline buffer solution in a substrate- independent coating manner. FT-IR and XPS spectra confirmed the successful formation of heparin-like coatings. Endothelial cell staining and morphological observations showed that heparin mimic coating can significantly promote cell adhesion and proliferation. In addition, systematic in vitro studies on coagulation, protein adsorption, platelet adhesion, and blood-related complement activation have shown that heparin-mimic macromolecule-coated substrates can significantly inhibit thrombogenic potential and inflammation induced by the material interface. DA-g-SAS was applied onto Fe3O4 nanoparticles. SAS-coated Fe3O4 nanoparticles can combine the advantages of magnetic responsiveness and blood compatibility. The measurement results show that the modified nanoparticles show improved anticoagulant properties and good recyclable properties. This study provides a promising method for introducing nanomaterials into the field of hemodialysis. Fe3O4 nanoparticles modified by heparin mimicking polysaccharide biomacromolecules can be considered as attractive materials for potential hemodialysis applications.
