Hericium erinaceus ethanol extracts inhibits inflammation and oxidative stress of brain on Japanese encephalitis virus infected mice

摘  要: Japanese encephalitis virus (JEV), a neurotropic flavivirus, is one of the major causes of acute encephalitis in human. The mortality range between 5 to 60 %. However, it often results in severe neurological sequelae and learning disabilities after rehabilitation. Hericenones and erinacines were isolated from the fruiting body and mycelium of Hericium erinaceus, respectively, and most of the compounds promote NGF biosynthesis in rodent cultured astrocytes. Oxidative stress and inflammation in neuron-glia system are key factors in the pathogenesis of neurodegenerative diseases. The aims of this research are to explore the neuroprotective and anti-inflammatory activities against hydrogen peroxide (H2O2) and LPS by Hericium erinaceus ethanol (HEe) extracts. In chemical analysis, HEe (1mg/mL) extract’s total phenol content is 24.48 mg/g, it also has great antioxidant activities on DPPH radical scavenging assay and the ferrous ion chelating assay. In vitro, when the concentration of HEe is 20 μg/mL, it can’t inhibit the JEV to infect SH-SY5Y and BHK-21 cells. In oxidative stress and inflammation assay, HEe can’t against the oxidative stress in cells, but it can reduce 13% nitric oxide (NO) production which compare with LPS. These results demonstrate HEe have potential efficacy to inhibit the LPS-stimulated inflammation on RAW 264.7 cells.
