The effect of bacteriophage on biofilms and its enhanced ability when combined with other antibiotics

摘  要: Biofilm is one of major causes of foodborne illness. A biofilm is defined as a sessile microbial community in which microorganisms live attached to a surface in a highly hydrated extracellular matrix. The presence of the extracellular matrix and the heterogenicity of the cellular metabolic status make biofilm-embedded bacteria up to 1000 times more resistant to most antimicrobial agents than their planktonic counterparts. The challenge of biofilm treatment is to investigate alternative strategies for its eradication. In this study, bacteriophage is chosen based on the administration of viruses selectively infecting target bacteria. The first reference aims to study the effect of bacteriophage on STEC O145:H25, and further test the effect of reducing transfer from stainless steel to beef. Results show that bacteriophage was effective in inactivating STEC O145:H25 in solution and in biofilms, and it can further reduce the number of STEC O145:H25 transferred from stainless steel to beef. However, it cannot eradicate all bacteria, which results in the recover of the bacteria. Therefore, in the next reference, we try to find the solution to eradicate all bacteria. In this content, the author tried the bacteriophage in combination with different regular antibiotics. Results show that bacteriophage Sb-1 can eradicate the MRSA biofilms, but it has its limitation. However, in the results of combinations of bacteriophage and antibiotics, the best concentrations are 105 27 PFU/mL bacteriophage in combination with 64 μg/mL rifampin 28 and 32 μg/mL daptomycin, which can eradicate the biofilm totally. If use bacteriophage
29 as pre-treatment, followed by different antibiotics, can also enhance its ability to totally 30 eradicate the biofilms.
