Aiming toward applications in the biomedical, hydrogels need to be stable at physiological pH with low degradation rates to maintain their three-dimensional porous structures. (Facchi et al., 2017) In addition, antibacterial drug-loaded hydrogel dressings can also realize sustained release of the drugs and achieve the corresponding long term antibacterial effect. In this study, a new type of composite wound dressing hydrogel aiming at avoiding wound infection was developed through embedding drug loaded gellan gum microspheres into a doubly crosslinked hydrogel, Therefore create durable gellan hydrogels, by associating anionic gellan with chitosan or schiff base reaction , avoiding conventional chemical crosslinking agents to produce polysaccharide-based hydrogels. After being embedded with cargo-loaded gellan gum microsphere, the gelation time, swelling index, degradation rate and mechanical properties of the blank hydrogel was improved, and the sustained release of cargoes (antibacterial drugs, tetracycline hydrochloride and silver sulfadiazine) was observed. Moreover, the excellent antibacterial activity of the composite hydrogel was also demonstrated in vitro. These results support the bioactive composite hydrogel can be employed as a promising injectable scaffold for promoting wound regeneration and drug delivery.
