Characterization of two cold adapted alginate lyases from different sources

Alginate oligosaccharides (AOS) produced by enzymatic degradation show versatile physiological functions and biological activities. Cold-adapted alginate lyases have unique advantages for alginate oligosaccharide preparation. In this study, two alginate lyases from different sources were investigated. First alginate lyase AlyS02 was discovered from Flavobacterium sp. S02 and second alginate lyase Alyw201 was discovered from Vibrio sp. W2. Two recombinant alginate lyases were respectively expressed in Yarrowia lipolytica. The optimal reaction temperature and pH of AlyS02 were 30 °C and 7.6. AlyS02 contained more than 90% enzyme activity at 25 °C. AlyS02 exhibited bifunctional activity to release disaccharides and trisaccharides from alginate. The molecular weight of Alyw201 is about 38.0 kDa, and the specific activity is 876.4 U/mg. The optimal reaction temperature is 30°C. It has more than 70% activity in a wide pH range of 5.0-10.0, Alyw201 is also NaCl-independent and salt-tolerant, and the degradation product is an oligosaccharide with DP (degree of polymerization) 2-6. These findings suggest that AlyS02 and Alyw201 would be a potent tool for the industrial applications.
