Effect of epigallocatechin-3-gallate (EGCG) on acute lung injury: inhibit inflammatory signal transduction

摘  要: Acute lung injury (ALI)/acute respiratory distress syndrome (ARDS) are serious clinical complications with a high frequency of morbidity and mortality. The initiation and amplification of inflammation is a well-known aspect in the pathogenesis of ALI and related disorders. Epigallocatechin-3-gallate (EGCG), a major constituent of green tea, has been shown to have protective effects on oxidative damage and has been shown to have an anti-inflammatory effect. The activation of inflammation is well- recognized as a vital factor in the pathogenesis of lipopolysaccharide (LPS)-induced ALI. Therefore, suppression of the inflammatory response could be an ideal strategy to prevent ALI. The aim of the study was to explore whether EGCG alleviates inflammation in sepsis-related ALI. Combining the above results, EGCG reduced LPS- induced ALI as it modulates macrophage polarisation towards M2 status, and also reduced the expression of proinflammatory M1 mediators iNOS TNF-α, IL-1β and IL- 6 in the LPS administered lung microenvironment, it increased the expression of KLF4, Arg1 and ym1, known to augment the M2 phenotype of macrophages. Therefore, EGCG attenuated LPS-induced ALI as it decreased the changes in blood gases and reduced the histological lesions, wet-to-dry weight ratios, and myeloperoxidase activity.
