Anti-obesity and Antidiabetic Effect of Sargassum on Obese and Type 2 Diabetic Mice Model

Obesity is a serious health problem worldwide and can lead to the development of metabolic syndromes, including hypertension, type 2 diabetes, and hyperglycemia. Simultaneously, the seaweed industry has attracted much attention in development of functional materials for food, cosmeceutical, nutraceutical, and pharmaceutical industries. And among the seaweeds, brown algae have revealed healthy effect because of its bioactive compound. Therefore, this study aims to investigate the anti-obesity and antidiabetic effect of Sargassum on mice model, including C57BL/6 and streptozotocin20 nicotinamide (STZ-NA) induced type 2 diabetes mice model. To identify whether
Sargassum horneri (SH) has an anti-obesity effect, C57BL/6 mice fed with high fat diet containing SH (100 and 300 mg/kg) were investigated for 7 weeks; for antidiabetic effect, STZ-NAinduced type 2 diabetes mice were gavaged daily with fucoxanthin (400 mg/kg), either free or encapsulated into porous starch (PS), and metformin (50 mg/kg) for three weeks. Then, the body weight, lipid profile, and histological analysis have been implemented. The results of SH increased the mRNA expression of mitochondrial uncoupling protein 1 and mitochondrial uncoupling protein 3 in brown adipose tissues that led to reducing body weight, white adipose tissue weight, fat accumulation in liver. On the other hand, both free and encapsulated fucoxanthin could decrease the fasting blood glucose and increase the plasma insulin level similar to metformin (p < 0.05), also significantly prevented the weight gain ( p < 0.05 ). Moreover, the histopathology evaluation of pancreatic tissue in diabetic mice exhibited that administration of metformin and fucoxanthin caused regeneration of pancreatic beta cells. These results showed that the ethanol extract of SH not only has anti-obesity effect but also antidiabetic effect and fucoxanthin is indeed retained antidiabetic toward type 2 diabetes. Besides, the healthy effect of Sargassum extract as a suitable bioactive supplement which can be used in functional foods for natural chronic diseases therapy.
